personalizat de terapie fotodinamic\u0103 (PDT) utiliz\u00e2nd fotosensibilizatori porfirinici inovativi \u0219i modulatori redox, formula\u021bi inteligent pentru tratamentul leziunilor cutanate premaligne, cum ar fi cheratoza actinica (AK). Proiectul se \u00eencadreaz\u0103 \u00een tematica Bioeconomie \u2013 Biotehnologie.<\/p><p>Aplica\u021bia medical\u0103 va fi demonstrat\u0103 utiliz\u00e2nd celule \u0219i modele de \u0219oareci. Se va investiga at\u00e2t eficacitatea terapeutic\u0103, c\u00e2t \u0219i aspectele toxicologice ale noului protocol PDT. Vom \u00eencepe proiectul la nivelul TLR 2 \u2013 formularea tehnologiei\/conceptului de produs, baz\u00e2ndu-ne pe un portofoliu de \u0219ase brevete \u0219i expertiza echipei \u00een domeniul PDT \u0219i al medicinei redox. Vom avansa prin TRL 3 \u2013 validarea conceptului experimental, prin studii in vitro si ex vivo utiliz\u00e2nd linii celulare \u0219i explanturi de piele de la \u0219oarece. Din aceste considerente dezvoltarea de strategii inovative privind diseminarea m\u0103surilor de preventie \u0219i furnizarea de substan\u021be active cu poten\u021bial antitumoral sunt esen\u021biale pentru combaterea cancerului la nivel mondial. Leziunile non-melanom (keratoze actinice, carcinom spinocelular \u0219i bazocelular, limfom cutanat, sarcom Kaposi, angiosarcom) sunt printre cele mai frecvente forme de cancer. Dezvoltarea unui cancer non-melanom este determinat\u0103 de factori genetici, istoric de infec\u021bie HPV, istoric de afec\u021biuni cutanate inflamatorii cronice, expunere la substan\u021be toxice, la soare, imunosupresie, fototipurile cutanate I \u0219i II, PUVA ca terapie anterioar\u0103 [3, 4]. Keratozele actinice sunt descrise in literatura sub forma de ,,<em>carcinom in situ<\/em>\u201d (din cauza keratinocitelor displazice asem\u0103n\u0103toare cu carcinomul cu celule scuamoase) sau ca leziune premalign\u0103 [5]. Abordarea terapeutica \u00een cazul keratozei actinice trebuie s\u0103 \u021binteasca sc\u0103derea riscului de malignizare \u0219i un diagnostic precoce al forma\u021biunilor maligne care pot ap\u0103rea \u00eentr-un camp de keratoz\u0103 actinic\u0103. Porfirinele, prin profilul lor structural si spectral, au potential \u00een identificarea \u0219i terapia manifest\u0103rilor cutanate de tip malign [6]. \u00cen plus, avantajul major al acestor tipuri structurale asem\u0103n\u0103toare hemului este selectivitatea pentru celule tumorale \u0219i versatilitatea lor, posibilitatea model\u0103rii structurale prin ata\u0219are de substituen\u021bi cu diferite grade de polaritate, pentru obtinerea unui raport hidrofil\/lipofil optim pentru internalizarea celular\u0103.<\/span><\/p><p><span style=\"color: #000000;\">In acest context, <strong>obiectivul principal<\/strong> al proiectului este de a elabora un protocol pentru terapia fotodinamica (PDT) a keratozei actinice, utilizand noi derivati porfiriniici. Activitatile din cadrul proiectului includ evaluarea potentialului de marker si agent antitumoral al unor compusi porfirinici cu arhitectura moleculara asimetrica si profil spectral adecvat. O prim\u0103 etap\u0103 a proiectului prevede ob\u021binerea de noi structuri porfirinice, prin tehnici ecologice de sintez\u0103 care respect\u0103a cerin\u021bele actuale din domeniul sintezei medicamentului. Corelarea datelor ob\u021binute a confirmat structurile compu\u0219ilor porfirinici sintetiza\u021bi \u00een aceast\u0103 etap\u0103. Concluzion\u00e2nd, studiile preliminare au condus la obtinerea, cu randamente bune, a 4 compusi porfirinici prin abordarea unei metode de sinteza moderne si ecologice.<\/span><\/p>\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-05f9c90 elementor-widget elementor-widget-text-editor\" data-id=\"05f9c90\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"text-editor.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t\t\t\t\t<p><span style=\"color: #000000;\">\u00cen <strong>Activitatea I.2<\/strong> au fost analizate 3 tipuri de formul\u0103ri pentru aplica\u021bii topice ale compu\u0219ilor porfirinici \u0219i modulatorilor, \u00eentro prim\u0103 etap\u0103 fiind selec\u021bionat\u0103 \u0219i detaliat\u0103 metoda \u00een gel.<\/span><\/p>\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-afce350 elementor-widget elementor-widget-text-editor\" data-id=\"afce350\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"text-editor.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t\t\t\t\t<p><span style=\"color: #000000;\">Studiul <em>in vitro<\/em> realizat \u00een <strong>Activitatea I.3<\/strong> a ar\u0103tat c\u0103 to\u021bi compu\u0219ii porfirinici analiza\u021bi, evalua\u021bi <em>in vitro<\/em> pe celule normale \u0219i tumorale specifice pielii (keratinocite umane normale HaCaT, fibrobl\u0219sti umani dermici HS27 \u0219i celule de mealnom de \u0219oarece B16F10) au avut o bun\u0103 biocompatibilitate cu celulele normale \u0219i tumorale de piele. Se remarc\u0103 totu\u0219i urm\u0103toarele aspecte: 1) compusul P5.2 poate inhiba multiplicarea keratinocitelor, ceea ce are relevan\u021b\u0103 terapeutic\u0103 \u00een cazul keratozei actinice caracterizat\u0103 prin hiper-proliferarea acestor celule, dac\u0103 se evit\u0103 fotosensibilizarea temporar\u0103 a pielii; b) compu\u0219ii P2.1 \u0219i P5.2, dar nu \u0219i derivatul porfirinic P4.2, determin\u0103 cre\u0219terea eliber\u0103rii LDH de c\u0103tre fibrobla\u0219tii dermici, posibil datorit\u0103 unor efecte nedorite care perturb\u0103 integritatea membranar\u0103 a fibrobla\u0219tilor dermici.<\/span><\/p><p><span style=\"color: #000000;\">To\u021bi compu\u0219ii porfirinici analiza\u021bi s-au incorporat \u00een celulele normale \u0219i tumorale de piele investigate, este adev\u0103rat cu intensit\u0103\u021bi diferite, dovedind astfel propriet\u0103\u021bi de marcator fluorescent.
